Brentuximab vedotin (Adcetris)
Brentuximab vedotin is een anti-CD30 ADC geregistreerd voor Hodgkin-lymfoom en systemisch anaplastisch grootcellig lymfoom. In de eerstelijns gecombineerd met AVD (A+AVD) bij gevorderd stadium cHL (ECHELON-1): PFS-verbetering versus ABVD. Bijwerkingen: perifere neuropathie, neutropenie.

Payload: MMAE (microtubulus-remmer). Hodgkin-lymfoom: A+AVD als eerstelijns bij gevorderd stadium (ECHELON-1 — PFS superieur aan ABVD). Monotherapie bij recidief na auto-SCT. Consolidatie na auto-SCT bij hoog risico (AETHERA). ALCL: eerstelijns (A+CHP, ECHELON-2). Bijwerkingen: perifere neuropathie (cumulatief, dosislimiterend), neutropenie. G-CSF profylaxe aanbevolen bij A+AVD.
